From Preface: For me, history has always moved out and away in concentric circles from my family's farm, the place I know best, to Longwood Township, to Pettis County, to Missouri, and finally to the United States. I have always felt that history should be taught the same way; that is, local history, should be taught first so that students will understand that what happned at the county seat in any given year may have been the result of decisions made and forces unleashed far away in the capitals of states and countries or in the boardrooms of corporations in Chicago or New York. For example, decisions made a thousand miles away in Washington, Dr. C. and Richmond, Virginia, ultimately resulted in the 1862 murder of a wealthy Pettis County land and slave owner accused of giving aid and comfort to the Rebels. Heath's Creek is where that unfortunate gentleman lived and where he was shot in cold blood by Union troops. There as well, where a few government barns and houses remain, is where Franklin Roosevelt's New Deal expermiented with cooperative farming in an effort to ease the plight of dispossessed Depression farmers. Let us not talk about battles in Virginia and dams on the Tennessee without first talking about skirmishes and WPA projects at home. Then let us move on to the bigger picture. Events did ot just happen in Pettis County; men made them happen. I adhere to the idea that history is generally made up of the personal histories of men and women. A History of Northeast Pettis County is more often than not the stories of the individual people who at one time or another lived there. Versions of some of the stories are told in other books; the reaminder have somehow been overlooked by historians. Yet in their day these now-forgotten men and women played important roles in the county, region, and state. The northeast quarter of Pettis County has had more than its share of those who helped open the West in the years before the War between the States. It was also home to others who made Pettis a leading and progressive agricutlural county after that war. The scope of this book includes the history of Heath's Creek, Longwood, Hughesville, Cedar, and Bowling Green Townships, and does not include Sedalia. Pettis County offers a rich heritage to all of its citizens, narrative or not, but to be proud of this legacy we must first know it. The purpose of the manuscripts was not to describe the recent experimental findings from each laboratory as is done in most conventional manuscripts. Instead, the purpose of the articles found within this focused volume of Molecular and Cellular Biochemistry was to describe how the technique is performed on the laboratory bench so that others less familiar with the technique may be able to use it in their own labs. The subjects described in this volume can be generally subdivided into three categories: molecular biology, cell biology and basic biochemistry. The methods cover wide areas including various DNA and RNA expression technologies, transfection techniques, quantification of ion flux movement, measurements of lipid metabolism, advances in the culture of specific cardiovascular cell populations, and the use of confocal microscopy to examine cell structure and function. We thank all of the authors who have contributed so much of their time and efforts and, most importantly, shared the `secrets' of these valuable techniques with the rest of the cardiovascular research community.
